操作流程：

1.  從(cong) 室溫平衡60min後的鋁箔袋中取出所需板條，剩餘(yu) 板條用自封袋密封放回4℃。

2.  設置標準品孔和樣本孔,標準品孔各加不同濃度的標準品50μL；

3.  待測樣本孔先加待測樣本10μL，再加樣本稀釋液40μL；

4.  隨後標準品孔和樣本孔中每孔加入辣根過氧化物酶（HRP）標記的檢測抗體(ti) 100μL，用封板膜封住反應孔，37℃水浴鍋或恒溫箱溫育60min。

5.  棄去液體(ti) ，吸水紙上拍幹，每孔加滿洗滌液，靜置1min，甩去洗滌液，吸水紙上拍幹，如此重複洗板5次（也可用洗板機洗板）。

6.  每孔加入底物A、B各50μL，37℃避光孵育15min。

7.  每孔加入終止液50μL，15min內(nei) ，在450nm波長處測定各孔的OD值。

收集標本前必須清楚要檢測的成份是否足夠穩定。對收集後當天進行檢測的標本，儲(chu) 存在4℃備用，如有特殊原因需要周期收集標本，將標本及時分裝後放在-20℃或-70℃條件下保存。避免反複凍融。標本2-8℃可保存48小時，-20℃可保存1個(ge) 月。-70度可保存6個(ge) 月。部分激素類標本需添加抑肽酶。
